About this test

Purpose of the Test

  • Diagnose Infections: Identifies if a bacterial infection is present.
  • Guide Treatment: Helps determine the best antibiotic, as gram-positive and gram-negative bacteria respond to different medicines.
  • Identify Type/Shape: Determines if bacteria are cocci (spherical) or bacilli (rod-shaped).

Gram-Positive vs. Gram-Negative

  • Gram-Positive (Purple): Retain the primary crystal violet stain due to a thick peptidoglycan cell wall. Examples include Staphylococcus aureus.
  • Gram-Negative (Pink/Red): Do not retain the stain and take up the counterstain (safranin) due to a thinner cell wall and outer membrane. Examples include E. coli.

Procedure

1. Sample Collection: A swab, fluid, or tissue sample is taken from the site of infection.

When is it Needed?

Doctors order this test for symptoms like fever, fatigue, or infections in the lungs (pneumonia), throat, urine, or wounds.

Limitations

While fast, the test may not identify the exact species of bacteria and may not work for bacteria with atypical cell walls.
